Astragalus Ameliorates Endothelial Dependent Vasodilatation Dysfunction in Obesity Rat

Subjective To investigate whether the endothelial dysfunction of obesity SD rat which is caused by high fat diet can be ameliorated by acute and chronic astragalus intervention.Methods 20 male SD rats were divided into 4 groups.Rats were fed with normal diet for 4 consecutive weeks(control group,n=5),were given high fat diet for 4 consecutive weeks(fat group,n=5),were given astragalus through gastric perfusion and fed with high fat diet for 4 consecutive weeks(astragalus acute intervention group,n=5) and were fed with high fat diet for 4 consecutive weeks,and then were transfused with astragalus for 4 hours through external jugular vein(astragalus chronic intervention group,n=5).All rats were killed and arteriae aorta were placed in the organbath and connected to a force trranducer and amplifier.Then acethylcholine and sodium nitroprusside of increasing concentrations were added to relax the rings to record the endothelial dependent vasodilatation(EDV) and endothelium independent vasodilatation(EIV).Results The endothelial dependent vasodilatation was btter than the control group,significant difference of endothelium independent vasodilatation was not observed in all groups.Conclusions endothelial dependent vasodilatation was impaired by high fat diet and can be improved partially by acute and chronic astragalus intervention.
