Effects of Carboxymethyl Chitosan Thermosensitive Hydrogel on the Attachment and Proliferation of Human Periodontal Ligament Cells

Objective This study aimed to provide a theoretical basis of the carboxymethyl chitosan thermosensitive hydrogel in the field of periodontal tissue regeneration,by observing the cultured cells proliferation in the extracts of carboxymethyl chitosan thermosensitive hydrogel.Methods Primary and passage human periodontal ligament cells were employed to co-culture the extracts as 2%、10%、15%、20%、50% concentrations from the carboxymethyl chitosan thermosensitive hydrogel with the temperature sensitivity.The MTT assay were used to exam the the effect of different concentration extracts on the proliferation of human periodontal ligament cells for 48h、72h、96h.Results After 48h,the OD values of 15% group was higher than that of other experimental groups and control one;After72h、96h,the OD values of experimental groups were higher than that of control one,the OD values of 15% group was higher than that of other experimental groups;compare with the OD values of the other four experimental groups,the OD values of 10%、20% groups were higher than that of 2%、50% group(P0.05).Conclusion Carboxymethyl chitosan thermosensitive hydrogel have the facilitating role in the proliferation of human periodontal ligament cells,the best promotion effect is observed with the extracts concentration 15%,which will be used in clinical practice.
