Effects of Carboxymethyl Chitosan Thermosensitive Hydrogel on the Differentiation of Human Periodontal Ligament Cells

Objective This study aimed to provide a theoretical basis of the carboxymethyl chitosan thermosensitive hydrogel in the field of periodontal tissue regeneration,by putting the carboxymethyl chitosan thermosensitive hydrogel on the human periodontal ligament cells,then observing the cultured cells alkaline phosphatase activity to determine the optimal concentration.Methods Primary and passage human periodontal ligament cells were employed to co-culture the extracts as 2%、10%、15%、20%、50% concentrations from the carboxymethyl chitosan thermosensitive hydrogel with the temperature sensitivity.The enzyme kinetics methods were used to exam the the effect of different concentration extracts on the differentiation of human periodontal ligament cells.Results There were no statistical significance of the OD values between the experimental groups and the control one in 24hduration(P0.05).After 48h、72h,all of the OD values of experimental groups were higher than the control(P0.05).Furthermore,15%extracts group was the highest of OD values,there was statistical significance between different extracts groups(P0.05)while no statistical signification between the 15% extracts group and 20%、50% extracts groups(P0.05).Conclusion Carboxymethyl chitosan thermosensitive hydrogel have the facilitating role in the differentiation of human periodontal ligament cells,the best promotion effect is observed with the extracts concentration15%,which will be used in clinical practice.
