Effects of Hyperbaric Oxygen Treatment on the Neurological Functional Improvement and Expression of BDNF in Acute Traumatic Brain Injury Rats

Objective This study was made to investigate the effects of hyperbaric oxygen(HBO)treatment on the neurological function improvement and expression of BDNF in acute traumatic brain injury(TBI)rats.MethodsAdult Sprague Dawley rats were divided into three groups:sham-operated group,TBI group and HBO treatment group.The sham-operated group was subjected to skull open only but not brain injury;TBI rats were subjected to TBI procedure according to Alice method;the HBO treatment group was performed for HBO treatment for 10 days after TBI.The neurological severity scores(NSS)records were evaluated after 14 days post injury.Then all rats were sacrificed,and motor cortex was obtained.The expression of BDNF mRNA was determined with reverse transcription-polymerase chain reaction(RT-PCR).BDNF located in cytoplasma of neurons was evaluated by immunohistochemical staining.Results NSS records in the HBO group were significantly decreased at 14 day in HBO treatment group,compared to that in the TBI group.Also,the expression of BDNF mRNA and protein in the HBO treatment group was significantly up-regulated.Conclusions HBO treatment can significantly improve neurological function in rats with acute brain injury and increased BDNF expressions.
