STUDY ON THE SCREENING PHARMACON OF nptⅡAND THEIR EUIFABLE CONCENTRATION DURING PEANUT(ARACHIS HYPOGAEA L.)TISSUE CULTURE AND SEEDLING PERIOD

The experiment is consisted of two parts,tissue culture exanunafion and seedling period examina- tion ,trying to inspoct peanut sensitivity to kanamycin,neomycin and G418 which are screening pharamacon of ntpⅡ.Shoot and young leaf are used in the peanut tissue culture expehment.Two peanuts varieties,Fenghua 1 and Fenghua 2,are chesed for the test.The results are as follows:When the concentration of kanamycin is 400mg/L, or neomycin is 500mg/L,or G418 is 25 mg/L,shoots have great difficulty to grow up and the color of them be- come yellow or brown.When the concentration of kanamycin is 100mg/l,or neomycin is 300mg/l,or G418 is 10mg/l,young leaves cant differentiate to callus.Five concentration grads of kanamycin that are 0,0.1,1.0,5. 0,10.0g/L,were setted up in the test of seedling peried,daubing young fold leaves with different concenttation of kanamycin when young plant has three euphylla.The result is that when the concentration of kanamycin is 5g/ L,untransgenefic young leaves appear big and clear yellow spots.
