PGE_2 inhibits LPS induced chemokine MIP by DCs of mice in vivo

Objective:To study whether or not PGE2 inhibits LPS-induced inflammatory chemokine MIP-1α and MIP-1β by peritoneal DCs of mice in vivo.Methods: Mice were i.p.injected with LPS and PGE2.Peritoneal exudates were collected and subjected to ELISA assay for MIP-1α and MIP-1β.In addition,the peritoneal cells were stained with PE-CD11c antibody,then stained with rabbit anti-MIP-1α and FITC labeled anti-rabbit IgG,followed by FACS assay for cell surface marker CD11c and intracellular MIP-1α.Results: PGE2 inhibits MIP-1α and MIP-1β production in peritoneal fluids.PGE2 also inhibits the accumulation of DCs in peritoneal cavity and inhibits MIP-1α in single DCs.These inhibitory effects were mediated through EP2 and EP4 receptors.Conclusion: PGE2 inhibits DCs function in vivo,suggesting an anti-inflammatory function.
