Effect of human neutrophil α-defensin on the adherence of Klebsiella pneumoniae to lung epithelial cells

Objective:To investigate the airway defense mechanism against bacterial infection and evaluate the effect of human neutrophil α-defensin on the adherence of pathogens to lung epithelial cells.Methods:A549 cells were incubated with two clinical isolates klebsiella pneumoniae(kp03183,kp03116)in the presence of human neutrophil α-defensin at a concentration of 20μg·ml-1 for 4 hours.The number of bacteria adherent to cells was quantified by colony counting method.Results:Human neutrophil α-defensin caused a 12-fold increase of kp03183 and a 5-fold increase of kp03116 in the number of bacteria adherent to A549 cells respectively.Conclusion:Human neutrophil α-defensins have the ability to enhance the adherence of klebsiella pneumoniae to lung epithelial cells which might be beneficial for the clearance of bacteria from the respiratory tract.
