Effect of rat mesenchymal stem cells on lymphocyte proliferation in vitro

Objective: To investigate the effect of rat mesenchymal stem cells(MSCs) on lymphocyte proliferation.Methods: MSCs and lymphocytes were isolated and cultured from rat routinely.With 4 passages,MSCs were collected and irradiated with 60 Co-radiation.The irradiated MSCs and lymphocytes were co-cultured for 72 hours,and then lymphocyte proliferation was measured by MTT assay.Hoechst 33258 staining was used to identify cell apoptosis.Results: After co-culture with MSCs,lymphocyte proliferation was inhibited substantially(p0.05) and nuclear fragments were found.Conclusion: MSCs can inhibit lymphocyte proliferation,mechanism of which may be related with cell apoptosis.
