Preparation of the monoclonal antibody against vascular endothelial cell growth factor-A receptor(KDR)

Objective: To prepare the specific monoclonal antibody(mAb) against a cytoplasmic domain of vascular endothelial cell growth factor-A receptor(KDR).Methods: GST-KDR fusion protein was used to immune 8~10 weeks female BALB/c mice and prepare hybridoma cell lines which could secrete anti-KDR mAb by the hybridoma technology.Results: One hybridoma cell line named as 3E9B2G11 was successfully obtained.Conclusion: One hybridoma cell line has been successfully prepared to secret anti-KDR mAb with high specification and affinity.It supply the possibility that it could be rebuilt the mAb into a small molecule tyrosine kinase receptor inhibitor and moved into cytoplastic domain to inhibit the biological functions of VEGF-A as soon as it interacts with KDR.
