Study on mechanism of parthenolide induced apoptosis in human bladder cancer cells BIU-87

Objective:To investigate the effects and the possible mechanism of parthenolide on the proliferation and apoptosis of BIU-87 cells.Methods:Human bladder cancer cells BIU-87 were treated by parthenolide in different concentrations(0,10,20and40μmol·L-1).MTT assay was applied to determine the effect of parthenolide on proliferation of BIU-87 cells,while apoptosis was analyzed by flow cytometry.Next,the mRNA levels of bcl-2and baxin BIU-87 cells were analyzed by RT-PCR.Results:After parthenolide treatment,parthenolide could inhibit the growth and proliferation of BIU-87 cells,with increased apoptotic rates by flow cytometry(P0.05).The results from RT-PCR suggested that parthenolide treatment contributed to the reduced bcl-2mRNA level and the elevated bax mRNA expression in BIU-87cells(P0.05).Conclusions:Parthenolide could significantly inhibit the proliferation of human bladder cancer cells BIU-87 by induction of apoptosis,which may be associated with inhibiting bcl-2mRNA expression and activating bax mRNA expression.
