Screening of Improved Culture Medium for Exobasidium vexans

Tea blister blight disease is caused by Exobasidium vexans Massee.The pathogen grows so slowly on the PDA medium so that it affects the subsequent research on the fungus. By adding four different carbon sources and four different nitrogen sources,16 media were screened to determine the most suitable nutrient composition of the pathogen. In addition,the optimum ratio of nutrients for the growth of E.vexans was found by orthogonal test.The results showed that theeffect of carbon sources on the growth of E. vexanswas greater than that of nitrogen sources. Organic nitrogen is more suitable for its growth than inorganic nitrogen. The optimum nutrient was sucrose and yeast. The best medium was sucrose 25 g/L,yeast 12 g/L and potato 150g/L.Compared with the PDA medium,the growth rate of the pathogen on the optimum medium was faster and its hyphae was fuller.
