The Study of Genes Expressions Related to Inflammation in Two Rat Models of Traumatic Deep Vein Thrombosis

Through comparing the expression changes of genes related to inflammation in the acute and subacute traumatic deep vein thrombosis(TDVT) rat models,to initially explore the significance of inflammation response in TDVT.In acute and subacute TDVT models,each 150 SD rats were divided into the control and experimental groups.Directly clamping femoral vein combined with hip spica fixation was used to establish acute TDVT model and quantitative beating on bilateral posterior limbs combined with hip spica cast fixation was used to establish subacute model.According to the different biological states,the experimental rats were divided into 7 subgroups: the control,the post-traumatic instant,the initial period of thrombosis,the crest-time of thrombosis,thrombi solution,thrombi insolution and no thrombosis subgroups.Genechip Rat Genome 430 2.0 gene chips were applied to detect the RNA expression of femoral vein dynamicly.Through the fold change analysis,the differential expression genes were selected for pathway analysis.The expression changes of genes related to inflammation in the two models were compared and analyzed.Results showed that: in the two mod els,28 genes in total related to inflammation response presented differential expression(23 in acute model,24 in subacute model,19 in both of them,8 only in one).But the difference of expression levels between the two groups had a statistical significance(Z=-2.513,P=0.0120.05).The change of genes related to inflammation is correlated to TDVT,and which can through influence the function of endothelial cell to contribute to thrombosis.
