Simultaneous determination of seven major components in Hawthorn leaves extracts by HPLC

OBJECTIVETo establish an HPLC method to determine simultaneouly glucosylvitexin,rhamnosylvitexin,rutin,vitexin,hyperoside,ursolic acid and quercetin in Hawthorn leaves extracts.METHODSThe Agilent ZORBAX SB-C18 column(5μm,4.6×250mm,series number:880975-902) was used at the temperature of 40℃;The mobile phase consisted of A:1% HAc,B:CH3CN;gradient elution profile was 0~5min,A:95%~90%,B:5%~10%;5~15min,A:90%~80%,B:10%~20%;15~35min,A:80%~0%,B:20%~100%.The flow rate was 1.0mL·min-1.The DAD detector wavelength was set at 360nm during 0~20min,then 220nm during 20~30min,and 360nm during 30~45min.The reference wavelength was 385nm.RESULTSThe average recoveries of Ⅰ~Ⅶ,were 99.87%,99.86%,101.23%,100.65%,101.58%,101.45% and 98.37% with RSD of 1.26%,1.28%,1.35%,1.49%,1.32%,1.51% and 1.44%(n=6),respectively.CONCLUSIONThe method is simple,rapid,accurate and reliable.It can be used to control quality of Hawthorn Leaves extracts.
