ENZYME-LINKED IMMUNOSORBENT ASSAY(ELISA) FOR MICROALBUMINURIA AND ITS CLINICAL USE

Periodic monitoring of microalbumin uria could be used to detect diabetic nephro-pathy at early, potentially reversible state. The presence of urinary Alb might also be an early sensitive indicator of renal vascular injury due to high blood pressure. We developed a method for ELISA of microalbuminuria. A high titer anti-human Alb serum,anti(h Alb)IgG-HRP conjugate and anti(h Alb)IgG coated microplate were all developed in our laboratory. The method was sensitive and simple. The assay range was 5-500ng/ml and sensitivity was 0.76ng of albumin. The intraassay and inter-assay CV% were 6.4% and 16.4% respectively. Albumin excretion values were 8.86 (SD 9.23)mg/24h in healthy subjects (n = 80) and 40.4(SD 40.4)mg/24h in diabetics (n= 16). Albumin excretion values were 15.17(SD 14.42)mg/24h,21.46(SD 24.34)mg/ 24h and 24.6(SD 18.9)mg/24h in patients with essential hypertension of I, II, III stages (WHO criteria) respectively.
