Influence of GM-CSF and TNF upon the Anti tumor Activities of Human Dendritic Cells

The effects of GM CSF and TNF upon the anti tumor activities of freshly isolated dendritic cells in human peripheral blood(DC 0) and those cells cultivated for 36 hours(DC 36) were investigated in vitro Experiments were performed with a mixture of human hepatoma cell line BEL 7402,lymphokine and PHA activated killer cells(LPAK) and dendritic cells(DC 0 or DC 36) in the presence of GM CSF(500u/ml or 100u/ml) or TNF(5000u/ml or 500u/ml) The experimental results showed that the cytotoxic activity was obviously enhanced after addition of GM CSF with the maximal effect observed with 100u/ml;while in case of TNF addition,the cytotoxic activity was also increased,but the effect was concentration dependent In comparison with the DC 0 and DC 36 dendritic cells,the cytotoxic activity of the latter were remakably heightened This effect indicates that the cultivated dendritic cells have a stronger cytotoxic activity than the freshly isolated ones under the cooperative effect with GM CSF and TNF
