Effects of thyrostatic treatment on T,B lymphocytes and expression of costimulatory molecules on pripheral blood lymphocytes of patients with Graves' disease

The aim of this study is to investigate the effect of thyrostatic treatment on T,B lymphocytes and the expression of costimulatory molecules on peripheral blood lymphocytes(PBL) of patients with Graves' disease(GD),so as to explore its role on pathogenesis and development of GD in which 20 untreated GD patients,11 treated patients and 16 sex-matched healthy subjects were selected for study,and the expressions of CD3,CD4,CD8,CD19,CD40,CD80,HLA-DR and CD28 on PBL were analyzed by flow cytometry.The expression of CD19,CD19CD40,CD19HLA-DR and CD80 on the PBL of GD patients increased remarkably(P0.01 or P0.05) compared with that of healthy controls.The number of CD3~(+)CD8~(+)cells decreased with an increase of the ratio of CD3~(+)CD4~(+)/CD3~(+)CD8~(+) cells(P0.05) in GD patients,while the percentage of CD3~(+)HLA-DR~(+)T cells was significantly higher in GD than that of healthy control(P0.05).The expression of CD80 returned to normal value when patients were treated with thioureas,methimazole or propylthiouracil for 2 to 4 months.The percentages of CD3~(+)CD8~(+) and CD8~(+)CD28~(+) cells decreased and CD3~(+)HLA-DR~(+),CD4~(+)HLA-DR~(+) cells increased remarkably compared with that of healthy control.Yet,the ratio of CD3~(+)CD4~(+)/CD3~(+)CD8~(+) also increased in treated and remitted patients.These results indicate that there are more activated T lymphocytes in PBL of GD patients and humoral immunity in GD patients shows in a higher level compared with healthy control.The changes of the detected molecules are not always in accordance with the improvement of clinical symptoms in remitted patients.
