IFN-γ and IL-10 upregulate the expression of B-cell activating factor in human HL-60 cells

It has recently been found that B-cell activating factor(BAFF) were produced in excess in patients with various B-cell lymphomas or multiple myelomas.It is uncertain whether the pathologic conditions,for example,pro-inflammatory cytokines,influence the expression of BAFF in myeloid tumor cells.To explore the effect of cytokines including interferon-γ,interleukin-10 and interlekin-4 on BAFF expression in human HL-60 cells,a myeloid tumor cell lines,the cultured human HL-60 cells were treated with cytokines including IFN-γ,IL-10 and IL-4 for 1~3 days.The expression of membrane-bound BAFF on HL-60 cells was examined by flow cytometry analysis,the amount of soluble BAFF was detected by ELISA assay,and the level of BAFF mRNA was tested by real-time PCR method.The results showed that the expression of membrane-bound BAFF,soluble BAFF and BAFF mRNA in human HL-60 cells were significantly elevated(P0.05) after incubated with interferon-γ and interleukin-10.More importantly,the level of BAFF mRNA in HL-60 cells showed a maximal level of induction after 1 day of stimulation with IFN-γ and IL-10.The enhancement of membrane-bound and soluble form of BAFF by IFN-γ and IL-10 treatment was time-dependent,reaching a maximal value after a 2 days exposure to the cytokines.In addition,a marked reduction in constitutive amount of released BAFF following IL-4 treatment was observed(P0.05).However,IL-4 treatment did not change the levels of endogenous BAFF transcripts and cell-bound BAFF in HL-60 cells.These data suggest that IFN-γ and IL-10 might up-regulate BAFF protein levels through a transcriptional or posttranscriptional mechanism,whereas IL-4 treatment inhibits BAFF secretion in HL-60 cells.
