Identification and localization of melatonin receptor in skeletal muscle of human beings and its molecular features

Objective To detect and localize the melatonin receptor and its mRNA expression in human skeletal muscle. Methods The human skeletal muscle was obtained from healthy volunteers.Immunohistochemistry and reverse transcription polymerase chain reaction(RT PCR) were used to detect MR in human skeletal muscle and to identify its subcellular distribution. RT PCR product was detected by DNA sequencing.Results ①Electrophoresis of RT PCR product showed positive band of mt 1 subtype in human skeletal muscle . RT PCR analysis identified the existence of mt1 mRNA in the cells investigated. Sequencing of RT PCR product showed its DNA sequence coincided with the sequence from Genebank. ②The subtype of MR, mt 1 was detected to exist on the membrane, in the cytosol and nucleus of human skeletal muscle cells with immunohistochemistry. Conclusions The subtype of MR, mt 1 exists in human skeletal muscle, and distributes on the membrane, in the cytosol and nucleus. Hence skeletal muscle is the target organ of melatonin.
