Construction of ubiquitinated vaccine against type 2 herpes simplex virus

Objective To construct the DNA vaccine against type 2 herpes simplex virus(HSV-2) modified by ubiquitinated infectious cell protein 27(ICP27). Methods Ubiquitions and HSV-2 ICP27 377-459 gene sequences obtained by PCR amplification were fused. The fused gene was identified and then cloned into eukaryotic expression vector pcDNA3. 1, for obtaining recombinant vector UB-pcDNA3. 1 which was comfirmed by DNA sequencing. Results DNA sequence analysis showed the rate of homology was 100% comparing with the sequence from Genebank. Conclusions The DNA vaccine against HSV-2 modified by ubiquitination of ICP27 has been successfully constructed.
