Comparative study of the effect of exercise and estrogen on the bone mass and bone remodeling in ovariectomized rats

Objective To study the effect of exercise and estrogen on bone mass and bone remodeling in ovariectomized rats,and to explore the differences between exercise and estrogen in the prevention and treatment of postmenopausal osteoporosis.Methods A total of 48 female SD rats,5 months old,were randomly divided into Basal,Sham,Ovx(ovariectomized),Ovx+e(ovariectomized and estrogen),Ovx+run(ovariectomized and running) and Ovx+run+e groups.Diethylstilbestrol(0.025 mg·kg-1·d-1) was subcutaneously injected in Ovx+e and Ovx+run+e groups one week after ovariectomy for 12 weeks,while the rats in Ovx+run and Ovx+run+e groups were designed to run in a special cage everyday(6.4 m/min,60 min/d) for 12 weeks.The femur bone mineral density(BMD) and bone histomorphometry indexes were measured at week 13.Results The BMD in Ovx+e group was significantly higher than that in Ovx group(P=0.029).The BMD in Ovx+run group was increased significantly when compared with groups Ovx and Ovx+e(P0.05).The BMD in Ovx+run+e group was the highest among the 6 groups(P0.01).The percentage of bone trabecula area(%Tb.Ar) and the number of bone trabecula(Tb.N) in Ovx+e group were much more than those in Ovx group(P0.05),while the bone trabecula separation(Tb.Sp) in Ovx+e group was less than that in Ovx group(P0.01).The %Tb.Ar and Tb.N in Ovx+run group were significantly higher than those in Ovx group(P0.05),while significantly lower than those in the rest groups(P0.05).But Tb.Sp was opposite(P0.01).The %Tb.Ar and bone trabecula thickness(Tb.Th) in Ovx+run+e group were significantly higher than those in the rest ovariectomized groups(P0.05),but Tb.Sp in Ovx+run+e group was lower than that in Ovx and Ovx+run groups(P0.01).The percentage of perimeter(%L.Pm),bone formation rates(BFR/BS,BFR/BV,BFR/TV),the number of osteoclast(Oc.No),Oc.No/Pm and bone mineralization rate(MAR) in Ovx+e group were significantly lower than those in Ovx group(P0.05).The %L.Pm and BFR/BV in Ovx+run group were significantly lower than those in Ovx group(P0.01).The %L.Pm,BFR/BS,BFR/BV,BFR/TV,MAR,Oc.No and Oc.No/Pm in Ovx+run group were significantly higher than those in Ovx+e group(P0.01).The %L.Pm,BFR/BS,BFR/BV and BFR/TV in Ovx+run+e group were significantly higher than those in Ovx+e group(P0.01),while MAR,Oc.No and Oc.No/Pm in Ovx+run+e group were significantly lower than those in Ovx and Ovx+run groups(P0.01).It was shown that trabecular was well-arranged by exercise.Conclusion Both estrogen and exercise can improve the bone mass and suppress the high bone turn-over in ovariectomized rats.But the effect of estrogen to suppress the bone resorption and improve the bone mass is better than that of exercise,and exercise is more effective for promotion of MAR and bone formation.Estrogen combined with exercise has a synergetic effect.
