Establishment of injure model induced by colistin sulfate PC12 cells

This study aimed at constructing PC12 cells injury model in vitro,which could provide experimental carriers and platforms for screening the medicine protecting against colistin sulfate-induced neurotoxicity.PC12 cells were exposed to different concentration colistin sulfate,MTT assay were carried out to evaluate cell viability and confirm the injure extent of cells at different time points.The results showed that except 62.5 μg/mL colistin sulfate presented the effect of promoting growth,other concentrations had a dose-dependent and time-dependent effect on injuring PC12 cells.The results indicated that PC12 cells with a cell density of 1×105cell per well incubated in 96-well plate for 24 h,and injured by 125 μg/mL colistin sulfate for 24 h,the reliable injure model of colistin sulfate in PC12 cell was successfully created.
