Preparation of typeⅡ collagen and its effect on the Rheumatoid Arthritis in rats

Objective:To obtain the type II collagen with higher purity and study its effect on the Rheumatoid Arthritis in rats; Methods:Type II collagen was digested to remove the telopeptide and purified using DEAE-Sepharose CL 6B ion exchange chromatography. Then, SDS-PAGE, amino acid analysis and X-ray were applied to identify the purity and structure of type II collagen. The rheumatoid arthritis model (CIA) was established in rats and CIA rats were treated by oral administration of type II collagen with different concentration; Results: The type II collagen with higher purity can be attained with typical triple helical structure. Oral administration of type II collagen obviously relieved the arthritis symptoms and markly suppressed the lymphocyte proliferation and IL-2. In addition, the levels of TNF-α and IL-2 in the experimental rats were markedly lowered versus vehicle-treated rats; Conclusion: Type II collagen could be applied to prevent and cure the RA as a functional healthy food.
