A Rat Liver Cell Line:Its Establishment and Some Characteristics Studies

A normal liver Cell line (NRL-84) derived from the new- born Wistar rat has been established in Vitro.The initial medium for cultivation was arginine-free Eagle's MEM supplemented with 10% fetal Calf serum until the 3th passages.Later it was changed to RPMI-1640 medium for continuous cultivation for more than one year (passed through 20 transfers).The cells are polygonal epithelial-like in morphology. Chromosome analysis showed a mode around 40—44 (68% frequency).AFP of this cells in the cytoplasm was assayed by immuno-fluorescent and in- situ nucleic acid hybridization methods.Counterimmunoelectrophoresis was adopted for the AFP detection in the old medium.The results showed that AFP was present both in the cytoplasm and in the medium and it gradually became weak or negative thereafter. Being the first of its kind reported in China,the establishment of normal newborn rat liver cell line will provide a satisfactory experimen- tal model and method for further studies on the mechanism of occurrence and development of malignant tumors.
