Study on Analysing Methods of Low Molecular Peptides with SDS-Polyacrylamide Gel Electrophoresis

To study the methods of analysing low molecular polypeptides and to simplify the three layers of tricine(3L\|T)\|sodium dodecyl sulfate\|polyacrylamide gel electrophoresis(SDS\|PAGE),we developed a two layers of tricine(2L\|T)\|SDS\|PAGE.Meanwhile,we also compared the differential function between 6mol/L urea and 24% glycerol in a 16% T,6% C separating arcylamide gel.Molecular weight markers(2\^512~16\^949kD) and samples to be measured formed obvious bands with one layer of a 8% T, 3% C acrylamide stacking gel.Molecular weight markers were analysed by linear regression(lg Mr \|μ) in the above 2L\|T\|SDS\|PAGE.These results suggest that low molecular weight polypeptides can be seen well both in the separating gel with 6mol/L urea and in the separating gel with 24% glycerol by 2L\|T\|SDS\|PAGE.The correlation coefficients of molecular weight markers are -0\^996 in urea and -0\^964 in glycerol.They are conventional,stable,simple to operate and are well established methods for analysis of low molecular weight polypeptides.\;
