STUDIES ON THE SECONDARY FORMATION OF PLASMODESMATA BETWEEN THE POLLEN MOTHER CELLS OF LILY BEFORE CYTOMIXIS

The transmission and analytical electronmicroscopy and the lead deposits procedurehave been used for ultrastructural studies andintracellular localization of acid phosphatasein PMCs of lily before cytomixis.The re-sults are summaried as follows: (1) The bydrolytic enzymes for the sec-ondary formation of plasmodesmata are se-creted via lysosomal-like vesicles or directlyfrom endoplasmic reticulum. (2) It is probable that hydrolysis forthe secondary formation of plasmodesmatamay be initiated on both sides of a wall(Figs.6,3).or perforation is initiated fromthe cell on only one side of the wall and iscompleted from that side (Figs.4,5,and 7). (3) Cytochemical acid phosphatase acti-vity is localised in lysosomal-like vesicles,endoplasmic reticulum and on the plasma membrane,which are corresponding to the elec-tron-dense material in transmission electronmicrograph. (4) X-ray microanalysis indicates tbatacid phosphatase reaction deposits are mostlikely lead phosphate.
