SOMATIC EMBRYOGENESIS AND PLANT REGENERATION FROM PROTOPLASTS AND EXPLANTS OF SEEDLINGS OF CROWNVETCH (FORAGE LEGUME)

Crownvetch (Coronilla varia L.) seedswere soaked in 98% sulphuric acid solutionfor 20—30 min,washed with sterile water(3changes),and inoculated on sucrose (0.3%)-agar(0.6%) medium (A-S). Protoplasts were released by EM-5 fromcrownvetch seedling cotyledons,yield being1.6±0.6x10~4/Pair.Protoplasts divided toform colonies in Kao's protoplast culture me-dium.division efficiency being 13±0.6%Somatic embryogenesis and plant regenerationwere induced from protoplast-derived calli onMSD~4. Callus formation,somatic embryogenesisand plant regeneration were induced fromcrownvetch seedling explants.roots,hypoco-tyls and cotyledons on MS-1,It was interes-ted in that many embryoids originated fromroots of explant-derived calli. Sections of specimens for microscopyeshow-ed that the plants were regenerated throughembryoids which originated from meristematictissues at the periphery and/or the interior ofthe calli. Increase of regeneration efficiency of em-bryoids from crownvetch protoplasts and con-ditions suitable for the regeneration planttransplantation will be studied.
