COMPARISON OF THE HYBRIDIZATION PATTERNS OBTAINED FROM LABELLED RNA WITH SOUTHERN BLOTTED VARIOUS RESTRICTION FRAGMENTS OF DNAS FROM RAT LIVER AND HEPATOMA

RNAs including nuclear RNA,poly A~+nuclear RNA,poly A~-nuclear RNA,poly-somal RNA and ribosomal RNA were iso-lated from rat liver and rat hepatoma BERH-2and labelled with Na~(125)I,γ-(32)~P-ATP,or α-(32)~P-pCp respectively.The hybridization oflabelled probes with restricted DNA frag-ments was compared by Southern blot technique.The hybridization bands of (125)~I-labelled nu-clear RNA with 15 kbp,9 kbp and 2.4 kbpEcoRI fragment of rat liver DNA were stron-ger than those of corresponding bands ofhepatoma DNA.when the 3'end or 5'endlabelled probes were used instead of the (125)~I-labelled probes,almost no hybridization bandon 15 kbp position of hepatoma DNA wasobserved.However,the hybridization band on2,0 kbp EcoR I fragment of hepatoma DNAwas not observed in corresponding site of ratliver DNA.The hybridization of probes and2.2 kbp BamH I fragment of rat liver wasabout 3 times higher than that with corre-sponding DNA fragment of hepatoma.
