EFFECTS OF CYTOCHALASIN B ON THE DEVELOPMENT OF POLLEN MOTHER CELLS OF SECALE CEREALE L.

Wang Xin-yu (Cell Biology Laboratory,Lanzhou University)
The effects of cytochalasin B on the de-velopment and microscopic and submicroscopic structure of pollen mother cells (PMCs) of Secale cereale L.were studied by aid of an-ther culture in vitro. After the anthers explan-ted at leptotene or zygotene stage were cultu-red on white's medium supplemented with cy-tochalasin B (5-40μmol/L) for 24 hours,a part of them failed to develop, blocked at zy-gotene stage and the percentage of the anthers stopped at zygotene stage increased with the increasing of the concentration of cytochalasin B.Among the anthers effected by cytochala-sin B,the PMCs of most of them stayed still in duration of cytomixis or pre-cytomixis.Further electron mirroscopic observation found that the plasmodesmatal channels between PMCs of the anthers blocked at zygotene stage remained open and were filled with cytoplas-mic matrix and some organelles and that chro-matin was blocked at the opening of plasmo-desmatal channels. The microtubule systems in the channel and nearby them existed still. The possible role about cytochalasin B is discussed.
