THE RELATIONSHIP BETWEEN TRANSPORT SITE OF HPD AND PHOTODAMAGE REGION OF HUMAN GASTRIC CANCER CELLS IN CELL CYCLE

This experiment was carried out by using hematoporphyrin derivative (HPD) and red light to study the biological effect of HPD in cell cycle of human gastric low-differentiated mucous adenocarcinoma cells.We have obser-ved that the distribution and transport of HPD were dependent on cell cycle; the HPD was transported from membrane to cytoplasm in the G_1 phase and directly into various region of cytoplasm in the S and G_2 phase,while HPD was scattered in the nuclear region in the M phase. The phototoxicity of HPD on synchronized cells after treatment by HPD and red light was increased in S and G_1 phase of the cell cycle,while M phase cells were found to be least sensitive.We also found that there was close relationship between distribution of HPD and photodamaged region of cells in cell cycle.Conspicuously,the binding of HPD to nucleo-lus was rapid and selective,especially in S phase cells whose nucleoli were found to be very photosensitive.
