ULTRASTRUCTURAL STUDIES ON SODIUM AND POTASSIUM ION ACTIVE TRANSPORTATIONS IN THE RAT EPIDIDYMIS.

Using the ultrastractural technique,rat epi-didymis was fixed for 30 min in a mixture of 2% paraformaldehyde and 0.1% glutaraldehyde,para-nitrophenyphosphate(P-NPP) was used as substrate,lead citrate as capture and dimethyl sulfoxide (DMSO) as activator.Levamisole was added to inhibit the non-specific alkaline phosp-hatase.To identify the specificity of NPPascoua-bain,a Na-K-ATPase inhibitor,or K-free mixture were used.It is interesting to find that two kinds of electron dense reaction precipitates in-dicating NPPase activities were observed in the distal efferent duct and epididymis of the rat.One is K-dependent,ouabain-sensitive and le-vamisole-resistant K-NPPase which was found on the basal and lateral plasma membrane of the principal cell in the distal efferent ducts and epididymis.Another,K-independent,oua-bain-insensitive and levamisole-resistant NPP-ase,was observed on the Golgi apparatus and the endoplasmic reticulum in the principal cells of the epididymis.There are no electron dense reaction precipitates in the basal,halo and clear cells.After gossypol treatment,K-NPPase ac-tivity of the plasma membrane of the principal cells in the distal efferent duct was decreased,especially in the lateral plasma membranes of the principal cells,while the K-independent NPPase activity in the principal cell of cauda epididymidis was higher than that of the con-trol rats.Similar results were obtained when K-free mixture were used.Na-pump was lo-calized in the principal cells of the epididymis of rat.
