DNA SINGLE-STRAND BREAKAGE AND ITS REJOINING IN HELA CELLS CAUSED BY HYPOCRELLIN A PHOTOSENSITIZATION

A hydroxylapatite batch assay for quan-titation of cellular DNA damage in cultured HeLa cells produced by the photosensitizer Hypocrellin A was used,which is a perylo-quinone derivative isolated from one kind of parasitic fungus Hypocrella bambusae sace.Hypocrellin A photosensitization causes DNA strand breakage.γ rays induce about 2 times more DNA single strand breaks than exposuse to Hypocrellin A and light when the two methods of treatment are compared on the same level of cell survival.In both cases the fraction rate of repair for Hypocrellin A pho-tosensitized strand breaks is smaller than that for γ ray induced strand breaks at the equiva-lent level of strand breakage.Hydroxyurea,actinomycin D and sodium butyrate suppress the repair of Hypocrellin A photosensitized breaks more strongly than they suppress repair of γ ray induced breaks.The repair rate and inhibitor sensitivity findings together suggest biologically important difference in the chemi-cal nature of γ ray induced and photosensiti-zed strand damages.
