ISOLATION AND ENRICHMENT OF CFU-E FROM MOUSE FETAL LIVER BY AFFINITY CHROMATOGRAPHY ON COLUMN OF SEPHAROSE-TRANSFERRIN

The mouse fetal liver cells were separated by affinity chromatography on column of Se-pharose 4 B covalently linked with transferrin.Almost all of the CFU-S(81%) and CFU-GM (84%) were eluted with no enrichment; a very high proportion of BFU-E(79%) and virtual-ly all CFU-E(88.3%) were bound on the column and desorbed using transferrin (20 mg/ /ml) with a 6.3 and a 9.2-fold-enrichment.This result has shown that the transferrin re-ceptor is expressed on the majority of eryth-roid progenitors,but only weakly on myeloid progenitors,and the method may provide an approach to isolating CFU-E.
