EMBRYOGENESIS IN CELL SUSPENTION OF PEUCEDANUM TEREBINTHACEUM

Xia Guang-min and Chen Hui-min (Biology Department,Shandong University)
Embryogenic calli could be obtained from the seedling root of Peucedanum terebin-thaceum (Fisch).ex Turcz.(A Chinese me-dicinal herb),cultured on MS medium con-taining 0.5 mg/1 2,4-D. These calli were used for the establish-ment of suspension culture.Embryogensis in suspension culture was induced in the presence of 2,4-D,and the embryoids formed developed into plantlets when they were transferred to 2,4-D-free medium. Cyto-histological observations revealed that embryogenesis started with single cells, followed by the formation of filamentous str-uctures,cell aggregates, calli with embryo-genic cell clumps, primary somatic embryo and different stages of somatic embryo.Several pattern of development from filamentous struc-tures to cell aggregates were observed.Light microscope section showed that the embryoge-nic cells initiated not only at the surface but also in the internal of embryogenic calli.
