ISOLATION OF GIBBERELLIN-REGULATED cDNAs FROM RICE (ORYZA SAT1VA) BY MEANS OF DIFFERENTIAL DISPLAY

Gibberellins (GAs) is one group of the most important plant growth regulators throughout the whole plant development. A differential display cloning scheme for the isolation of differentially expressed cDNA was established, involving reverse transcription of total mRNA and polymerse chain reaction amplification of the cDNA subpopulation. From indica rice 15 differential cDNA fragments, which were subsequently recovered from gels and reamplified, had been identified by comparisons of amplified cDNA fragments between samples from control and test seedlings which had been treated with GA3 (one sort of GAs). One of the differential cDNA fragments, DDF1, was hybridized to total rice DNA and RNA, and it was confirmed that the corresponding gene of the cDNA is a single copy one whose high-level expression could be induced by high concentration of GA3.
