Isolation,identification and antibacterial activity of the actinomycetes from iron mine wasteland in Lveyang of Shaanxi

The study aims at isolation and identification of the actinomycetes in Iron Mine in Lveoyang of Shaanxi Province and attempts to study the antibacterial activity of actinomycetes metabolites. The soil samples have been collected and purified by tissue culture method in Lveyang Iron Mine,Shaanxi Province,and the initial row weight has been observed by colony morphology. All strains are sequenced by 16 S r RNA,and their genotypes are determined by constructing the developmental tree. Then in order to determine its genus by establishing phylogenetic tree,Escherichia coli,Staphylococcus aureus,Bacillus subtilis,Salmonella typhl and Canidia Albicans were selected as indicator bacteria when the antimicrobial activities of these actinomycetes were tested through antimicrobial experiments in exterior body. Through the colony morphology and microscopic observation of rescheduling,14 strains were identified,of which 10 strains belong to Streptomyces,3 strains belong to Micromonospora 1 strains belong to Promicromonospora. The result shows that 14 strains can produce antimicrobial substances,10 strains of actinomyces have more obvious antibacterial activity,accounting for71. 43% of the total. It is concluded that the tailing soil is rich in strain. By morphological and molecular identification,it can be concluded that Streptomyces belong to a large proportion of the isolated actinomycetes,and strains with strong antibacterial activity are screened out,which provides a theoretical basis for enriching and excavating actinomycete resource bank.
