A study of micro-quantitative methods for crystal fiber in herbs from glycyrrhiza uralensis

Objective:Improve the micro-quantitative methods for crystal fiber in herbs fromglycyrrhiza uralensis.Methods:Based on existing micro-quantitative methods,they replace"Shuifei"sample-making method(the process of which solid powder and liquid are grinded together in order to make suspension or remove contaminants)with Suspending sample-making method,and substitute blood-counting plates for common microscope slides.Besides,we determine sample quantity by linear analysis and optimize the proportion of suspension which is used for plates making as well as the number of sample plates for micro-quantity with orthogonal experiment method.Through inspecting the suspension stability,they could confirm the precise time period of the experiment.Results:The sample fineness should be 100 mesh.We need 10 slides to determine the sample quantity.Suspension:The mix of 60% glycerol and choral hydrate in the ratio of 19∶1.The average number of micro-characteristics of 1mg glycyrrhiza uralensis come from Shenmu is(1498±22)/mg.Conclusion:It is a practical way to use blood-counting plates to increase the ease of operation and accuracy of micro-quantitative method.
