Experimental study on determination of phosphatidylcholine content in egg yolk by NMR

By using fresh egg liquid as the raw material and phosphatidylcholine as the research object,the ethanol(extraction)-acetone(degreasing,recrystallization)combined extraction method is used to extract and purify the yolk lecithin.The qualitative and quantitative analysis method for phosphatidylcholine is established by using the NMR technology with the triphenyl phosphate as internal standard.The experimental results show that when the ratio of the material to liquid is 1:10,the volume fraction of ethanol is 95%,the extraction time is 60 min,and the amount of the lecithin extracted from egg yolks is 4.88—8.14/g.The results of the NMR hydrogen spectrum and phosphorus spectrum show that the percentage of phosphatidylcholine in these yolk lecithin extracts ranges from 16.9%to 27.4%.The results of recovering the phosphatidylcholine from egg yolk by adding standard are satisfactory,which indicates that the method can be used for the determination of phosphatidylcholine in egg yolk.
