Effect of Bacillus Bifidus on Mucositis Induced by Methotrexate in Rats

Objective To explore the mechanism and therapeutic effects of bacillus bifidus on mucositis induced by Methotrexate (MTX) in Wistar rats.Methods Fifty-four Wistar rats were randomly divided into 3 groups.Group A was normal control group.All rats in group B and C received intraperitoneal injection of MTX 1 mL(100mg/kg).The rats in group C were fed with bacillus bifidus through mouth twice each day before the 2 days of MTX injection until executed.Rats were killed on day 1,3,5 after MTX injection.Then tumor necrosis factor-α (TNF-α) was detected on intestines by means of immunohistochemistry.Secreted immunoglobulin(sIgA) was detected on intestinal mucosal fluid by enzyme linked immunosorbent assay.Bacterial translocation from intestine was detected by bacterial culture of mesenteric lymph node.The change of tissue morphology and the mortality were observed.Results Compared with group B,the change of tissue morphology had improved significantly in group C,villus height and crypt depth were higher significantly on 1,3,5 day(Pa0.05); the expression of TNF-α in endothelial cell was decreased significantly (P0.05);sIgA in intestinal mucus increased significantly (P0.05),and bacterial translocation decreased significantly on 3,5 day(Pa0.05).In the end,it degraded the mortality(P0.05).Conclusions The MTX-induced mucositis is an acute impairing procedure.Bacillus bifidus can mitigate mucositis obviously,and shorten the course of disease.It provided a theoretical basis for the administration of bifidobacteria to prevent the side effects in chemotherapy.
