Effect of Chronic Hypoxia on Endogenous Adenosine Concentration in Plasma of Rats

Objective To observe the effect of chronic hypoxia on changes in plasma adenosine concentration and its mechanism in rats.Methods Forty-two male Sprague-Dawley rats were randomly divided into normoxic group(control group),hypoxia groups of 1 day,3 days,5 days,7 days,14 days,21 days groups respectively(H1 group,H3 group,H5 group,H7 group,H14 group,H21 group),and 6 rats were in each group.The levels of endogenous adenosine,adenosine deaminase(ADA) activity and plasma 5′-nucleotidase(5′-NT) contents were determined.SPSS 16.0 software was used for statistical analysis.Results After hypoxia,plasma adenosine content in rat began to increase on the first day,and it reached the peak on the third day,and there was a significant difference compared with the control group(P0.001),then with the longer hypoxia time,the concentration of plasma adenosine gradually decreased,on the 14th day and the 21th day,compared with control group,the differences were not significant compared with the control group.5′-NT plasma concentration level was significantly increased on the third day(P0.001),and it gradually declined the longer hypoxia time,but it was still higher than that in the control group(P0.001).ADA activity in plasma in the early stage of chronic hypoxia significantly increased(Pa0.001),with longer hypoxia time,the activity decreased slightly,but still was higher significantly than that in the control group(P0.001).Conclusions Chronic hypoxia in an early stage can lead to an increase in plasma adenosine,and it gradually decreases in a later stage,which is associated with 5′-NT levels and ADA activity.
