Studies on Methods for Purification and Titer Determination of Antitoxin against Pseudomonas Aeruginosa Exotoxin A(PEA)

Li Jingyu et al(Lanzhou institute of Biological Products, Lanzhou 730046)
This paper described the comparative studies of several main factors which probably influence the refining effect of equine anti-PEA(pseudomonas aeruginosa exotoxin A)plasma in the procedure for purification by pepsin treatment and ammonium sulfate precipitation. Meanwhile, the optimal assays measuring titer of PEA antitoxin were selected to obtain more accurate comparative parameters. The results revealed that it could be used to purification of PEA antitoxin equine plasma by pepsin treatment and ammonium sulfate precipitation, and the bioassays including neutralization of cytotoxicity and neutralization of mouse lethality were accurate for titer determination of PEA antitoxin.Based on the comparative results,a modified procedure for purification of PEA antitoxin equine plasma was recommended and compared with routine procedure. It was suggested that the efficiency of the purification by modified procedure was fine.
