Quantitation of the Vi Antigen in Salmonella Typhi Polysaccharide Vaccine by Rocket Immunoelectrophoresis

The Vi capusalar polysaccharide of Salmonella typhi is a linear homopolymer of a -1, 4-inked N-acetylgalactosaminuronic acid, variably O-acetylated at the C-3 position. It cannot be measured by conventional colormetric methods. It is demonstrated that logarithm of polysaccharide concertration is proportional to the height of the rocket. We detected the Vi concentration of 18 batches Salmonella typhi polysaccharide vaccine from six manufacturers by rocket immunoelectrophoresis. Most of the vaccines are in keep with the requirements,but some of them surpass the highest value which defermined by the requirement. The three batches of each manufacturer give similar results,this reflects that the working-tecnological process is stable. We can do the test very easly in a short time with small monunt of sample,so the rocket immunoelectrophoresis is a good method for quantitation of the Vi antigen in Samonella typhi polysaccharide vaccine.
