Response surface optimization of the medium components used for cultivation Paecilomyces hepialid

The response surface methodology was used to optimize submerged fermentation medium components,in order to improve yeild of mycelial adenosine content from Paecilomyces hepialid.Firstly,The Plackett-Burman Design was used to evaluate the effects of eight variables on adenosine content,and two factors including potato and silkworm powder were found to have significant effect on adenosine content of Paecilomyces hepialid.Then,the steepest ascent method was used to determine their optimal levels to lead the experimental values closely toward maximal response surface region.Finally,central composite design and response surface methodology were further adopted to derive a statistical model for optimizing the medium components for adenosine production of Paecilomyces hepialid.The experimental results indicated that the optimal medium components were: potato(8.25%),sucrose(1%),corn flour(1%),peptone(0.5%),silkworm powder(0.81%),KH2PO4(0.1%),MgSO4·7H2O(0.1%),(NH4)2SO4(0.1%),respectively.It is by using of the optimal medium components constituted medium,the adenosine content of Paecilomyces hepialid reached to 3.44 mg/g from original 2.15 mg/g.
