ISOLATION AND CHARACTERIZATION OF FLEXIBACTER CHINENSES SP. NOV.

Strain FCA was isolated from theCalothrix nursery bed in the Institute ofHydrobiology. It is a flexible rod or fila-ment in shape with uniform width andvarious length (0.4--0.5 by 4.0--16.0μm). When it is grown in the liquid me-dium the length is usually more than 60.0μm. Sometimes the cells aggregate to forma mass. Motile by gliding, resting stagesnot known. Gram-negative. The colour of colonies of strain FCA ispink, With a pale iridesence, and turn toorange with ageing. But on the third davthe cell mass becomes colourless. The ma-ximum absorption peak of crude extract ofpigment (in n-hexane or ethanol) is at 484nm. Facultatively anaerobic; hydrolyzesstarch; many sugars are utilized and acidis produced from glucose, maltose, lactose,galactose, sucrose, xylose, anabionose, raf-finose and cellobiose, but not from manni-tol, fructose and glycerol. Cellulose (fil-ter paper or carboxymethyl cellulose), chi-tin and agar are not attacked. Caseinhydrolysate, tryptone, ammonium or nitrateis reduced to nitrite. Gelatin and tyrosineare not degraded. The DNA G+C content of strain FCAis 34.2 moles%. The initial isolate possesses ability tolyse vegetative cells of Anabaena strain1105. Growth of strain FCA is inhibited by0.01% SDS, susceptible to tetracycline 10u/ml; resistant to 10 μg actinomycin D,2,000 u polymyxin B or E, 1,600 u penicil-lin G, 60 u streptomycin, 25 μg mitornycinC, 600 u erythromycin, 100 u chloramphe-nicol, and 200 u neomycin per ml. etc. This strain belongs to the genus Flexi-bacter, but it differs significantly fromspecies previously described. Therefore itis regarded as a new species, for which thename Fleribacter chinenses is proposed. Type culture FCA is deposited in the Ins-titute of Hydrobiology, Academia Sinica,Wuhan.
