TRANSFECTION OF RNA FROM μ_2 RNA PHAGE

Three methods: 1) Ca~(2+) treatment, 2)hypertonic medium, and 3) lysozyme-EDTAsystem in the presence of protamine sulfatewere applied to compare the efficiency oftransfection of purified μ_2 RNA. Amongthem, the third method is the most effec-tive for transfection with plaque formingunits (p. f. u) reaching to 2.1×10~6 per μgRNA. The action of protamine sulfate andother influencing factors on the transfectionof μ_2 RNA, such as the concentration ofRNA, the partial digestion of the RNA mo-lecule in 5' or 3' terminal ends, the integra-tion of ethdium bromide and actinomycinD, and the heat denaturation were also ob-served and discussed in detail.
