Mechanism of Parkinson's disease resulted from ubiquitin-proteasome system dysfunction

Parkinson's disease(PD) is a neurodegenerative disease.The proteasome inhibitor-induced PD can simulate the progression of human PD.The dopaminergic neurons in substantia nigra not only showed degeneration and loss but also displayed intracyto-plasmic Lewy body in the survival neurons,which is closely related to the pathological features of human PD.Therefore,ubiquitin-proteasome dysfunction has become the focus for PD study.This article reviewed the function of ubiquitin-proteasome system(UPS),the role of proteasome inhibitor Lactacystin in UPS and the mechanism of defects in dopaminergic neurons resulted from UPS dysfunction.
