In vitro plant regeneration from first stem internode explants of water spinach(Ipomoea aquatica)

In this experiment,white bone willow leaf spinach(Ipomoea aquatica Forsk.) was used as research material.By screening explants and hormone ratios,an in vitro regeneration system of I.aquatica from first stem internode explants was established for the first time.Results showed that a large number of sterile I.aquatica seedlings could be obtained by adding 0.05% PPM to the basic culture of MS.Explants of cotyledons and hypocotyls did not induce adventitious buds;however,the first stem internode successfully induced adventitious buds.The optimal medium was MS+1.0 mg/L 6-BA+0.1 mg/L IAA,and the adventitious bud induction rate was 20%.The optimal medium for inducing adventitious bud roots was 1/2 MS +0.1 mg/L NAA,with the rooting rate reaching 100%.The whole regenerated seedlings were transplanted into matrix soil,with a transplant survival rate of 100%.This study established a spinach regeneration system and laid the foundation for further research on transgenic I.aquatica.
