Effects of exogenous p16 gene on the cell cycle and growth of renal carcinoma cell

Objective To study the cell cycle and growth-inhibitory effects of exogenous p16 gene on renal carcinoma cell line. Methods A eukaryotic expression vector containing p16 gene was transfected into human GRC-1 renal carcinoma cells using lipofectamine to investigate the effect of exogenous p16 gene on the tumor cells. Results ①Immunohistochemistry revealed that p16 protein were significantly expressed in the transfected GRC-1 renal carcinoma cells. ②The measurements by flow cytometry proved that exogenous p16 gene mainly blocked the G 1 to S of the cycling of renal carcinoma cells. ③The growth rate of cells transfected with p16 gene was significantly inhibited when compared with the growth rate of pcDNA3-LacZ or PBS treated cells. Conclusion Exogenous p16 gene has growth-inhibitory effects on renal carcinoma cells with cycling specificity.
