Distribution and expression of KCNE2 in heart tissue from spontaneously hypertensive rats

Objective To investigate the distribution and expression of KCNE2 in different parts of myocardium of spontaneously hypertensive rats(SHR) and compare differences of KCNE expression in cardiac muscle between young and old SHR.Methods First membrane proteins were prepared from right ventricular myocardium(RV),interventricular septum,left ventricular myocardium(LV) and atrial myocardium from young and old SHR,respectively.Then the membrane proteins were separated by SDS-PAGE,transferred to polyscreen PVDF membrane,probed with a primary anti-KCNE2(polyclonal Ab),and visualized with ECL detection kit.Images were acquired and analyzed with related software.Results KCNE2 protein was found to distribute in all different parts of myocardium of SHR with no significant differences.The expression of KCNE2,however,in right ventricular and interventricular septum of old rats was significantly higher than that of young rats.Conclusion KCNE2 protein distributes in all different parts of myocardium of SHR.The expression of KCNE2 in right ventricular and interventricular septum of old rats is significantly higher than that of young rats.
