Establishment of CD4~+CD25~-CD45RB~(high) T cells transfer colitis model in mice

Objective To establish a chronic colitis model induced by CD4+CD25-CD45 RBhigh T cells and analyze its role in investigation of inflammatory bowel disease.Methods CD4+CD25-CD45 RBhigh T cells were sorted and purified from C57BL/6 mice spleen cells by flow cytometry,and then transferred intravenously into Rag1-/-mice.Body weight of each group of mice was recorded every week,and HE staining of colon sections was performed according to the standard protocol.Lamina propria CD4+T cells were isolated and TNF-αand IFN-γlevels in lamina propria CD4+T cells were examined by ELISA method.Results The Rag1-/-mice reconstructed with CD4+CD25-CD45 RBhigh T cells developed body weight loss at week3,followed by evident body weight loss at week 6,accompanied with diarrhea. HE staining results of colonic sections showed evident infiltration of inflammatory cells,presence of mucosal erosion and ulceration,and transmural inflammation spread to submucosal.TNF-αand IFN-γlevels in lamina propria CD4+T cells were significantly higher than those in the control group.Conclusion CD4+CD25-CD45 RBhigh T cells transfer colitis model plays an important role in the study on the pathogenesis of inflammatory bowel disease.
