Experimental Study on Identification of Salmonella Typhimurium Using Monoclonal Antibodies

We developed an ELISA method using monoclonal antibodies against Salmonella typhimurium ( McAb, STM) for detection of STM. 162 clinical isolates of STM were investigated by ELISA and staphylococcus protein A coagglutination test ( SPA- CoA) simultaneously, and a comparison was made between these two methods. The results indicated that the ELISA method was sensitive enough to detect 5×102/ml of STM, and its specificity was also high; the detection rate of STM 0-4 antigen is 100% while that of H-i antigen is 96.3%. Furthermore, this study revealed that the sensitivity and specificity of the McAbs used were all comparable to the current standard of the diagnostic serum factor for Salmonella. The experimental method showed a promising prospect of application.
